About GHRP-6 Acetate
GHRP-6 (Growth Hormone Releasing Hexapeptide) is a synthetic 6-amino acid peptide with the sequence His-D-Trp-Ala-Trp-D-Phe-Lys-NH₂ that acts as a potent agonist at the ghrelin receptor (GHS-R1a/growth hormone secretagogue receptor). The peptide incorporates D-amino acids at positions 2 (D-Trp) and 5 (D-Phe) for protease resistance, and the C-terminal lysine amide is essential for GHSR-1a binding. GHRP-6 was one of the first synthetic GH secretagogue peptides characterized pharmacologically and served as the structural lead for subsequent GHS-R1a agonist development, including ipamorelin.
In laboratory research, GHRP-6 is used extensively as a reference ghrelin receptor agonist for GHSR-1a pharmacological characterization, receptor binding affinity studies, downstream calcium mobilization and ERK1/2 signaling pathway analysis, and comparative potency determination between GH secretagogue peptide generations. Unlike ipamorelin, GHRP-6 also activates cortistatin and PACAP receptor systems at higher concentrations, making it a useful selectivity counterpart in receptor profiling studies.
