About SS-31
SS-31 (Szeto-Schiller peptide 31; elamipretide) is a synthetic tetrapeptide with the sequence D-Arg-2',6'-Dmt-Lys-Phe-NH₂, where Dmt represents the non-natural amino acid 2',6'-dimethyltyrosine. The alternating cationic and aromatic residues create an amphipathic structure that selectively partitions into the inner mitochondrial membrane at concentrations that do not disrupt membrane potential. The 2',6'-dimethyltyrosine modification prevents oxidative degradation of the tyrosine residue while maintaining its electron-scavenging properties. SS-31 has high selectivity for cardiolipin, the signature phospholipid of the inner mitochondrial membrane.
Research investigations of SS-31 have centered on its interaction with cardiolipin in model lipid bilayer systems and isolated mitochondria, its effects on mitochondrial cristae architecture in electron microscopy studies, ATP synthase activity characterization in mitochondrial preparations, and cytochrome c peroxidase inhibition assays.
